摘要: |
目的研究骨肉瘤端粒酶与化疗耐药、多药耐药基因(mdrl)以及细胞凋亡抑制基因(bcl-2)的相关性,探讨骨肉瘤端粒酶的表达在化疗耐药中的作用。方法49例骨肉瘤石蜡标本以原位杂交检测其端粒酶、mdrlmRNA的表达,以免疫组化(S-P法)检测其bcl-2蛋白的表达,结合临床分析骨肉瘤的端粒酶表达活性与化疗耐药以及与mdrl和bcl-2表达的关系。结果49例骨肉瘤端粒酶的表达活性与化疗耐药负相关,与bcl-2表达活性正相关.而与mdrl表达不相关。结论骨肉瘤的端粒酶与肿瘤化疗耐药有关,机制可能在于端粒酶与bcl-2间的相互协同作用。 |

Abstract: |
Objective To study the correlation among the telomerase activity and chemotherapy resistance and multidrug resistance gene (mdr-1) and bcl-2 in osteosarcoma, and to evaluate the role of the telomerase activity in osteosarcoma chemotherapy resistance. Methods By using in situ hybridization, the expression of telomerase and mdr-1 were observed in 49 paraffin sections of osteosarcoma. The expression of bcl-2 were also examined by using immunohistochemistry in this 49 paraffin sections. Furthermore, the relationships of telomerase activity and chemotherapy resistance and multidrug resistance gene (mdr-1)and bcl-2 were analyzed. Results The expression level of telomerase in osteosarcoma showed a negative correlation with chemotherapy resistance, and a positive correlation with the expression of bcl-2. There was no relation between the expression level of telomerase and the expression of mdrl. Conclusion The expression of telomerase is correlated with chemotherapy resistance, its mechanism may be laid in the synergism of telomerase with bcl-2. |
