Effect of phospholipase D1 on proliferation of pancreatic cancer cells

Abstract:
Objective To explore the role of PLD1 in regulating proliferation of pancreatic cancer cells. Methods The effects of PLD, PLD1 and PLD2 inhibitors on the cell viability, proliferation and cell cycle of pancreatic cancer were detected by CCK-8, EdU fluorescent staining and flow cytometry, respectively. Furthermore, the effects of PLD1 knockdown or overexpression on the cell viability, proliferation and cell cycle of Capan-2 cells were detected. Results Both PLD and PLD1 inhibitors inhibited the proliferation of Capan-2 cells(P=0.004,0.044), while PLD2 inhibitors did not have significant effects on the proliferation of Capan-2 cells(P=0.945). Downregulation of PLD1 decreased the cell viability(P=0.000), cell proliferation(P=0.002) and suppressed cell cycle progression of Capan-2 cell(P=0.004,0.002), while upregulation of PLD1 enhanced the cell viability(P=0.004), cell proliferation(P=0.002) and promoted cell cycle progression of Capan-2 cell(P=0.0004,0.035). Conclusion PLD1 promotes the proliferation of pancreatic cancer cells, which may be involved in the progression of pancreatic cancer.
