目的探讨健康围绝经期女性泌尿生殖道及肠道微生物群落结构。
方法纳入50名健康围绝经期女性,采集其口腔、尿液、肠道、宫颈、阴道上段及阴道下段6个部位标本,利用Illumina NextSeq2000测序平台对16S rRNA的V3-4可变区扩增产物测序,全面分析围绝经期女性泌尿生殖及肠道微生物群落结构特征。
结果在健康围绝经期女性中,不同部位菌群的丰度、均匀度和多样性存在差异。获得序列可归为41个门、82个纲、131个目、285个科、792个属及1889个种。其中有4个优势菌门: 厚壁菌门(Bacillota),放线菌门(Actinomycetota),拟杆菌门(Bacteroidota)和变形菌门(Pseudomonadota)和19个核心菌属。不同系统6个部位的菌群组成差异显著,在属水平,肠道部位丰度最高的菌属为芬戈尔德菌属(Finegoldia),口腔部位为链球菌属(Streptococcus),4个泌尿生殖道部位为乳杆菌属(Lactobacillus)。在种的水平,口腔部位丰度最高的菌种为口腔链球菌(Streptococcus oralis),肠道部位为大芬戈尔德菌(Finegoldia magna),泌尿生殖道部位丰度最高的菌种为惰性乳杆菌(Lactobacillus iners)和卷曲乳杆菌(Lactobacillus crispatus)。
结论通过对健康围绝经期女性泌尿生殖道及肠道微生物群落结构的分析,全面多系统描述了该阶段女性微生物组结构特征,为这一生理年龄阶段女性多发妇科疾病的诊治提供了新的微生物学视角参考。 |

Abstract: |
ObjectiveTo investigate the distribution of urogenital and gut mocrobiota in healthy perimenopausal women.
MethodsSpecimens from mouth, urine, gut, cervix, vaginal upper and lower segment were collected from 50 healthy perimenopausal women. The V3-4 variable region amplification products of 16S rRNA were sequenced with Illumina NextSeq2000 sequencing platform to analyze the characteristics of urogenital and intestinal microbiota.
ResultsThe abundance, evenness, and diversity of flora at various sites in healthy perimenopausal women were different. The obtained sequences included 41 phyla, 82 classes, 131 orders, 285 families, 792 genera and 1889 species. There are four dominant bacteria: Bacillota, Actinomycetota, Bacteroidota and Pseudomonadota, and 19 core bacteria genera. At the genus level, the bacterial genus with the highest abundance in the gut was Finegoldia, while the highest abundance in the mouth was Streptococcus. In the four urogenital tract sites, the highest abundance was Lactobacillus. At the species level, the highest abundance in the mouth was Streptococcus oralis, the highest abundance in the gut was Finegoldia magna, the highest abundance in the urogenital tract was Lactobacillus iners and Lactobacillus crispatus.
ConclusionThe analysis of the urogenital tract and gut microbiota in this study provides a comprehensive multi-system description of the microbial community structure characteristics in perimenopausal women, which may be conducive to the diagnosis and treatment of gynecological diseases common in women of this physiological age group. |
