营养性肥胖大鼠模型的建立及评价

摘要:
目的高脂饲料喂养诱导营养性肥胖大鼠模型的建立及评价。方法将45只成年sD大鼠随机分为正常模型组(n=40)和对照组(n=5),分别给与基础饲料和高脂饲料喂养8周,检测大鼠体重、体长、体重指数(bodymassindex,BMI)、血糖及总胆固醇(cholesterol,CHO)、甘油三酯(triglyceride,TG)、高密度脂蛋白(highdensity lipoprotein—cholesterol,HDL—C)及低密度脂蛋白(low density lipoprotein—cholesterol,LDL-C)。结果通过高脂饲料喂养后,模型组大鼠体重、BMI与对照组比较差别均有统计学意义(P〈0.05,P〈0.01);其总胆固醇及LDL—C明显高于对照组(P〈0.05,P〈0.01),HDL—C则明显低于对照组(P〈0.01),造模过程中,甘油三酯两组差别无统计学意义;模型组大鼠存在糖耐量异常。结论通过高脂饲料喂养可诱导出符合或贴近临床的营养性肥胖大鼠模型。

Abstract:
Objective To establish and evaluate the high -fat- induced obese rat model. Methods Forty-five adult Sprague-Dawley rats were randomized into model group (h=40) and normal group (n= 5) with high-fat diet and standard diet for 8 weeks, respectively. Measurements of body weight, body length, body mass index (BMI), blood glucose and total cholesterol (CHO), triglyceride (TG), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C) were performed. Results There were statistically significant differences in body weight and BMI between model group and normal group (P<0.0.5, P<0.01). The model group showed a significantly higher total cholesterol and LDL-C (P<0.0.5, P<0.01), and lower HDL-C than normal group(7J<0.01). There was no significant difference in triglyceride between 2 groups. Abnormal glucose tolerance was found in model group. Conclusion Feeding with high-fat diet can induce rat model with nutritional obesity which is accordant or close to clinic.
