急性胰腺炎大鼠血清IL-32变化及意义

摘要:
目的 观察急性胰腺炎(acute pancreatitis,AP)大鼠IL-32、IL-31、TNF-α和淀粉酶(amylase,AMY)的变化,探讨IL-32在大鼠急性胰腺炎中的变化及意义。方法 64只SD大鼠随机分成2组:AP组(急性胰腺炎组)和SO组(假手术组),每组分4个时间点(3 h、6 h、12 h、24 h),每个时间点8只大鼠。采用4%牛磺胆酸钠胰胆管逆行注射制备AP动物模型,SO组仅行开腹后翻动胰腺。在各组相应处理后3 h、6 h、12 h、24 h分别留取大鼠的胰腺组织及静脉血以全自动生化仪检测AMY,ELISA检测血清IL-32、IL-31、TNF-α表达,并作胰腺组织病理学检查。结果 AP组的AMY、TNF-α以及病理评分均比同时间点SO组显著升高(P<0.01);并在前6 h内随时间逐渐下降(P<0.05)。在AP发生3 h内,AP组IL-32比SO组显著升高(P<0.01),并呈显著上升趋势(P<0.05),且与TNF-α(r=0.899,P<0.05)及病理评分(r=0.782,P<0.05)呈正相关;6 h以后IL-32呈下降趋势(P<0.05),12h时降至与SO组相当。而IL-31在AP组与SO组之间和各时点之间均无差异(P>0.05),与TNF-α及病理评分也无相关性。结论 IL-32在AP早期上升,且与TNF-α及病理评分正相关,可能在AP早期起促炎因子的作用。

Abstract:
Objective To study the changes and significances of IL-32 in rats with acute pancreatitis(AP) by investigating the changes of IL-32,IL-31,TNF-α and AMY.Methods Sixty-four SD rats were randomly divided into AP group and sham operation(SO) group.Rats both groups were divided into 3h group,6h group,12h group,24h group with 8 in each.AP was induced in rats with retrograde infusion of 4% sodium taurocholate into the the pancreastobiliary duct(1ml/kg).Rats were sacrificed respectively 3,6,12 and 24 hrs after operation.Samples of blood serum and pancreas tissue were collected.AMY in blood serum was detected with automatic biochemistry analyzer.IL-32,IL-31 and TNF-α were determined by enzyme linked immunosorbent assay.The histopathology of pancreas tissues with HE staining were observed under the light microscope.Results 1.Levels of the AMY,TNF-α and pathological score in AP group were significantly higher than those in SO group at the same time(P<0.05),and all of them increased gradually with time in the first 6 hrs(P<0.05).The level of IL-32 in AP group was significantly higher than that in SO group within 3hrs(P<0.01).IL-32 was significantly positively correlated with TNF-a and pathological score within the first 6 hrs in AP group(r=0.899,P<0.05,r=0.782,P<0.05).IL-32 decreased in AP group 6 hrs later,with no significant difference from that in SO group in exactly 12 hrs.The level of IL-31 in AP group was no different from that in SO group(P>0.05) and was not correlated with TNF-α and pathological score at any time.Conclusion IL-32 increases at the early stage of AP and positively correlate with the TNF-α and pathological score.IL-32 may function as a pro-inflammatory factor in the earlier stage of AP.IL-31 has no change in the in the early stage of AP.
