目的研究V型肢原在气管移植后闭塞性气道疾病(obliterative airway disease, OAD)中的作用。方法V型肢原免疫C57BL/6小鼠,30d后接受C57BL/6小鼠原位气管移植,未接受移植的C57BL/6小鼠和同系移 植小鼠(C57BL/6→C57BL/6)作为对照。移植7 d后处死小鼠,移植气管H-E染色,观察移植气管气道病变情况。用RT-PCR和ELISA法检测移植气管相关炎症因子的含量。结果与正常对照组、同系移植组相比,胶原 免疫组移植气管气道闭塞程度明显增加。RT-PCR检测移植气管显示,肢原免疫组小鼠IL-1β、IL-6、IL-17A较正 常对照组及同系移植组有明显升高,ELISA法结果显示肢原免疫组小鼠EL>6较正常对照组及同系移植组有明显 升高。结论V型胶原免疫小鼠接受原位气管移植后,移植气道闭塞程度明显加重,这可能与V型胶原诱导的 自身免疫反应有关。 |

Abstract: |
Objective To study the role of collagen type V (ColV) in obliterative airway disease after tracheal transplantation.Methods Pathogen-free C57BL/6 mice were immunized with Col (V) and received syngeneic orthotopic tracheal transplantation 30 d latter. Untreated normal C57BL/6 mice and non-immunized syngeneic transplanted mice were used as controls. The transplanted tracheae were harvested after 7 d,stained with H &E,and examined under light microscopy. Inflammatory factors were detected by RT-PCR and ELISA.Results The occlusion of the transplanted tracheae were more severe in Col ( V) immunized mice compared with untreated normal mice and non-immunized syngeneic transplanted mice. The level of IL-1β, IL-6 and IL-17A were higher in Col( V) immunized mice compared with two control groups. Conclusio Severe tracheal occlusion develops after syngeneic orthotopic transplantation in Col(V) immunized mice, in which immune response may be involved after transplantation. |
