目的探讨在人肝星状细胞系中miR-27b基因表达异常对LX-2细胞的脂滴沉积与迁移能力的影响。方法转染干预miR-27b的表达,油红染色与划痕实验检测LX-2细胞的脂滴沉积与迁移能力,荧光定量PCR检测靶基因RXRa的表达。结果miR-27b在人肝星状细胞系LX-2细胞活化过程中表达升高,miR-27b的模拟物可以促进LX-2细胞的迁移、减小胞质内脂滴沉积;相反,miR-27b抑制物可以减弱LX-2细胞的迁移,增加胞质内脂滴沉积。荧光定量PCR结果显示RXRa在转染miR-27b模拟物表达降低,抑制物无明显变化。结论上调miR-27b的表达可能是通过靶基因RXRa促进LX-2细胞的迁移,减少细胞胞质内的脂滴沉积。 |

Abstract: |
Objective To investigate the expression of miR-27b in human hepatic stellate LX-2 cells and its effects on cell lipidosis and migration. Methods miR-27b mimic or inhibitor were transfected into LX-2 cells. The lipid droplets in cells was detected by oil red staining, cell migration was examined by scratch test and the expression of RXRa was detected by Real-Time PCR. Results The expression of miR-27b was up-regulated during the activation of LX-2 cells. After the transfection of miR-27b mimic, the migration of LX-2 cells was enhanced, while the intracellular lipid droplets were reduced and the expression RXRa was decreased. After the transfection of miR-27b inhibitor the migration LX-2 cells was reduced, while the intracellular lipid droplets were increased and the expression RXRa was not significantly changed. Conclusion The over-expression of miR-27b can enhance migration of LX-2 cells and reduce the intracellular lipidosis, which is probably through the target gene RXRa. |
