目的 比较五种不同的siRNA对进行性骨化性纤维增殖不良症模型细胞中ACVR1基因的抑制效果。方法 实验组分为A、B、C、D、E五组,分别转染五种不同序列的siRNA,对照组转染通用阴性siRNA和试剂。转染36h后,采用实时荧光定量PCR检验各实验组对ACVR1基因的抑制率。结果 与试剂对照组相比,只有E组观察到了25.5%的抑制率(P>0.05),其余各组未观察到抑制效果。结论 可以排除四种siRNA序列对ACVR1基因不具有抑制作用。转染正义链序列为5′-CCAGGUGGAUUGUUUCGAU-3′的siRNA对ACVR1基因可能有抑制作用。 |

Abstract: |
Objective To compare the effects of five different siRNAs on ACVR1 gene in fibrodysplasia ossificans progressiva (FOP) cells. Methods Five different siRNAs were trensfected into cultured FOP cells (groups A, B, C, D, E), non-trensfected FOP cells (negative control) and mock-transfected cells (mock transfection control) were used as controls. Real-time fluorescence quantitative PCR (RT-qPCR) was performed at 36h after transfection. Results Only a 25.5% suppression of ACVR1 was observed in group E (P>0.05). Conclusion siRNA with sense strand 5′-CCAGGUGGAUUGUUUCGAU-3′ may suppress the ACVR1 gene, while other 4 studied seque-nces are not effective. |
