目的 探究检测豚鼠咳嗽反应性的方法,为动物咳嗽反应性的评价提供方法学依据。方法 健康豚鼠及慢性咳嗽模型豚鼠雾化吸入不同浓度柠檬酸、辣椒素、肉桂醛溶液,记录单次吸入咳嗽次数及气道阻力相关指标。结果0.2mol/L柠檬酸、10-5mol/L辣椒素、20mmol/L肉桂醛作为咳嗽激发物时,与健康豚鼠相比,模型豚鼠咳嗽次数差异明显,而气道阻力无差异。联合刺激发现,不同激发物刺激咳嗽反应性存在个体差异。结论 0.2mol/L柠檬酸、10-5mol/L辣椒素和20mmol/L肉桂醛检测豚鼠咳嗽反应性最佳。选择多种激发物联合检测咳嗽反应性方法更好。 |

Abstract: |
Objective To investigate the optimal concentration of different challenge agents for testing cough reflex sensitivity in guinea pigs. Methods Unanesthesized healthy guinea pigs and guinea pigs of citric acid-induced chronic cough model were subjected to inhale citric acid, capsaicin orcinnamaldehyde with various concentrations. The cough counts and the data related to airway resistance were documented. Results When 0.2 mol/L citric acid, 10-5mol/L capsaicin or 20mmol/L cinnamaldehyde were applied as irritants separately, there were significant differences in cough counts between healthy guinea pigs and those with chronic cough, but no difference in airway resistance between two groups. Individual differences exited while the combination of different stimuli was applied. Conclusion 0.2mol/L citric acid, 10-5mol/L capsaicin or 20mmol/L cinnamaldehyde are the optimal concentration of challenge agents for cough provocation test in guinea pigs, and it would be better to use combination of various cough challenge agents. |
