杜仲叶提取物对人精子膜功能损伤的保护作用

摘要:
目的 探讨杜仲叶提取物对人精子膜功能氧化损伤的保护作用。方法 应用梯度离心法,分离出正常精子作为模型,在采用次黄嘌呤-黄嘌呤氧化酶体系产生活性氧(reactive oxygen species, ROS)环境下,使用维生素C(0.15mg/ml)及小、中、大剂量杜仲叶提取物(150、300、600mg/L)共同孵育精子悬液,检测精子膜中的丙二醛(malondialdehyde, MDA)含量和超氧化物歧化酶(superoxide dismutase, SOD)活力,通过精子尾部低渗膨胀试验、精子顶体完整率评估精子膜功能。结果 与模型组相比,维生素C和小、中、大剂量杜仲叶提取物在相同的条件下,均可提高精子悬液SOD活力,降低MDA含量(P<0.01)。与维生素C比较,小、中剂量的杜仲叶提取物对ROS所致精子膜的氧化损伤的保护作用差异无统计学意义(P>0.05),大剂量的杜仲叶提取物明显优于维生素C(P<0.01)。与模型组比较,小、中、大剂量杜仲叶提取物组的精子顶体完整率和精子尾部低渗膨胀试验结果均有统计学意义(P<0.01),大剂量杜仲叶提取物对精子的保护作用最显著(P<0.01)。结论 杜仲叶提取物对人精子膜结构和功能损伤具有显著的保护作用。

Abstract:
ObjectiveTo investigate the protective effect of extraction of Eucommia ulmoides oliv (eucommin) on the function of human sperm membrane under oxidative injury. Methods The normal sperms were isolated with percoll gradient centrifugation, and the reactive oxygen species (ROS) were produced with hypoxanthine-xanzine xanzine (HX-XO) system. The human sperms suspension in ROS environment was treated with vitamin C (0.15mg/ml) or different concentrations of eucommin (150,0 and 600mg/L). The malondialdehyde (MDA) content and superoxide dismutase (SOD) activity of sperm membranes were detected. The function of sperm membrane was analyzed by sperm hypotonic expansion test and the rate of intact acrosome was detected. Results Compared with oxidative model group, SOD activity of sperm suspension was improved,and the content of MDA was reduced in vitamin C group and eucommin groups (P<0.01); the effect of high-dose eucommin group was more marked than that of vitamin C group (P<0.01). There was significant difference in the rate of intact acrosome and sperm hypotonic expansion test between model group and eucommin groups (P<0.01), and the high-dose eucommin group showed better result than other groups (P<0.01). Conclusion Eucommin has a protective effect on human sperm membrane function from oxidative injury.
