环指蛋白6(ring finger protein 6, RNF6)是含有RING结构域的泛素连接酶E3,可作为骨架招募E2和靶蛋白,介导泛素由E2转移到底物特定位点。RNF6可通过泛素化修饰底物蛋白改变其表达水平,也可改变底物功能。RNF6参与SHP1/STAT3、Akt/mTOR、Wnt/β-catenin、ERα/Bcl-xL、MAPK/ERK、MAD1/c-myc等信号通路的调控,与多种恶性肿瘤的发生发展相关。靶向抑制RNF6表达或可作为抑制肿瘤发生发展的一种方法,但仍需大量研究。本文通过查阅近年来涉及泛素连接酶RNF6的文献,综述RNF6的发现历程、结构特征及其与恶性肿瘤的关系,旨在为后续深入研究RNF6对肿瘤的影响提供理论基础。 |

Abstract: |
As a ubiquitin-ligase E3 containing a RING finger domain, ring finger protein 6(RNF6) can serve as skeleton to recruit E2 and target proteins and mediate the transfer of ubiquitin from E2 to specific sites of the substrate. By ubiquitin modification, RNF6 can induce degradation of target protein or alter its function. RNF6 is involved in the regulation of a series of signaling pathways, including SHP1/STAT3, Akt/mTOR, ERα/Bcl-xL, Wnt/β-catenin,MAPK/ERK and MAD1/c-myc pathway. RNF6 is also associated with the occurrence and development of a variety of malignant tumors. Targeted inhibition of RNF6 expression may be used to suppress tumorigenesis and development, but further studies are still needed. In this article, recent research progress on the processes and structural characteristics of RNF6 as well as its relationship with maligmant tumors is reviewed to provide a reference for further studies. |
