miR-145在肝癌中的表达和功能研究

摘要:
目的探讨miR-145在肝癌组织、HepG2细胞株和肝脏正常组织中的表达情况及其对肝癌细胞株HepG2增殖和侵袭能力的影响。方法运用荧光定量PCR技术检测肝癌、HepG2细胞株及肝组织中miR-145的表达量.应用MTT法检测miR-145对HepG2细胞增殖的影响,通过侵袭实验观察miR-145对细胞侵袭能力的影响。结果肝癌组织中miR-145表达明显低于正常组织(P<0.05)。浓度为100 nmol/L的miR-145作用时间72 h时,对HepG2细胞的增殖抑制最强,侵袭实验显示100 nmol/L的miR-145 mimics处理组穿膜细胞数目明显减少,并与对照组有显著差异。结论miR-145在癌组织中的表达明显低于正常组织;miR-145抑制肝癌细胞株HepG2的增殖和侵袭能力,其在肿瘤发生发展过程中可能担任抑癌基因的角色。

Abstract:
Objective To investigate the expression of miR-145 in hepatic carcinoma,adjacent normal hepatic tissues and HepG2 cell line,and its effect on proliferation and invasion of human hepatic carcinoma.Methods RT-PCR was performed to determine the expression of miR-145 in hepatic carcinoma,adjacent normal liver tissues and human hepatocellular carcinoma HepG2 cells.Proliferation,invasion assays were conducted to examine the effects of miR-145 on HepG2 cells through up-regulating miR-145 expression by miR-145 mimics.Results Compared to normal hepatic tissues,miR-145 expression was decreased in liver cancer specimens and HepG2 cells(P<0.05).Proliferation assay revealed that 100 nmol/L miR-145 significantly inhibited HepG2 cell growth at 72h.A significantly different number of invaded HepG2 cells were observed between groups treated by 100 nmol/l miR-145 and controls.Conclusion The expression of miR-145 is decreased in hepatic carcinoma and HepG2 cell line.Up-regulation of miR-145 expression inhibits cell growth and invasion of HepG2 cells in vitro.
