摘要: |
目的对细胞培养过程中的支原体污染进行检测并探讨相应的预防措施。方法通过DNA荧光染色和聚合酶链反应(PCR)来检测细胞培养过程中的支原体污染,分别对有支原体污染细胞、未污染细胞、小牛血清及培养基进行了实验研究。结果支原体污染细胞能被Hoechst 33342染色且PCR产物在280 bp处有特异性阳性条带。结论通过两种实验方法联合检测为判断细胞是否为支原体污染提供准确依据,并能对支原体污染做出相应预防措施。 |

Abstract: |
Objective To investigate the methods for detection of mycoplasma contamination in cell culture.Methods Mycoplasma contamination was detected by DNA fluorescence staining and polymerase chain reaction(PCR) on mycoplasma contamination cells,uncontaminated cells,calf serum and culture media.Results Mycoplasma contamination cells can be stained by Hoechst 33342 and PCR assay showed specific positive bands at 280 bp.Conclusion Combination DNA fluorescence staining with PCR can effectively detect mycoplasma contamination in cell culture. |
